ISOLATION AND CHARACTERIZATION OF PLASTIC DEGRADING BACTERIA IN DUMP SITE IN JERE LOCAL GOVERNMENT AREA, MAIDUGURI, BORNO STATE
Abstract
This study "Isolation and Characterization of Plastic Degrading Bacteria in Dump Site in Jere Local Government Area, Maithiguri. Porno State. Plastic waste is a significant environmental problem due to its persistent nature and detrimental effects on ecosystems. Dump sites are known to harbor substantial amount ofplastic waste, making them important study sites for understanding the processes involved in plastic degradation. Plastic is a prevalent material used in daily life to make food package, furniture, decorating materials, and laboratory equipment. The aim of the study was to isolate and characterize bacteria from selected dump sites. Objectives 1. Identify and isolate plastic degrading bacterial from dump sites in Jere local government, Maidugurl Porno state. 2. Characterize the bacterial diversity. 3.To identifi physiological parameters of the dump sites on landfill. Samples were collected aseptically from 3 different dumpsites within Mara local governments, areas of Porno State. Samples were collected from 10-20 cm depth, samples were carried to Biology laboratory in a sterile container, the Soil Sample were processed asepticallyfor soil analysis and alsofor isolation ofbacteria. The soil sample was carried for weighing each sample was weigh 1gram before inoculating to prepared sabourad Dextrose Agar (SDA) by the process of serial dilution and incubated at 28 (room temperature) for 4-7 days or at 40 °c for 24 hours. Plastic degrading microorganisms were isolated from three different soil samples and degradations ofplastic strips by the isolated microorganisms was determined using contact to send dilution method. The isolation which shows high opacity was selected and further used The isolates obtained was subjected to standard biochemical test results showed the presence of Streptococcus spp., Bacillus spp. staphylococcus aureus. the organisms Identified was farther media and their bio degradative ability was observed stood determined by loss of weight after a period of 30days and observed that Backenal spp degrades up to 23 Ye. Thus it may take 120 days' complete degradation of plastics by using bacteria and its takes approximately 75 days while using fungi for complete degradation of plastic. Further biochemical test can be done to identifi the exact bacterial and fungal species. The study revealed that improper management of waste from generation point to the point of disposal account for bacterial contamination of soil. Aerobic bacterial are usually snore likely contaminates of soil than anaerobic bacteria, this is especially so considering the fact that the environment is aerobic. Necessary to isolate, identify and characterised Aalton in waste dumpsite and conduct a study over time, to form baseline information for I there is currently insufficient further studies as - client information on the health hazard caused • by waste dumpsite
