DETERMINATION OF LACCASE ACTIVITY IN DEGRADATION OF WASTE SACHET WATER BAG AS A POTENTIAL TOOL FOR BIOREMEDIATION
Abstract
With the development of industry and modern manufacturing, non-degradable low-density polyethylene (LDPE) has been widely used posing a rising environmental hazard to natural ecosystems and public health. This study focused on determination of biodegradation activity of laccase in Trichoderma harzianum against sachet water bag. The objective was to isolate and purify Trichoderma harzianum from soil and to determine laccase activity in Trichoderma against sachet water bag. Trichoderma harzianum was isolated and cultured on Potato Dextrose Agar (PDA), and laccase production was optimized. Enzyme activity assay was conducted to quantify laccase activity during the degradation process. Results showed that mean enzyme activity (in U/L) gradually increased with concentrations (in µg/). This study highlights a sustainable approach to managing plastic waste using microbial enzymes, contributing to eco-friendly waste management solutions.
