Abstract

ABSTRACT Siderophores are small organic molecules produced by microorganisms under iron-limiting conditions which enhance the uptake of iron to the microorganisms. They are synthesized and secreted by many bacteria, yeast, fungi and plants for Fe (iii) chelation. A variety of plants growth promoting bacteria (PGPR) colonize the rhizosphere and contribute to iron assimilation by plants. This study aimed to isolate and identify siderophore producing bacteria from rhizosphere.Rhizosphere soil sample was collected from botanical garden, bacterial load was determined through several dilution and enumeration. Characterization was conducted to identify the isolates, using Chrome Azurol Sulfonate assay (CAS) the identify isolate were screened for siderophore production. Bacterial colonies ranged from 2× to 5× with no colonies obtained from the control . A total of 5 siderophore producing bacteria were identified the rhizosphere soil which comprises of Azotobacter Bacillus subtilis, Esherichia coli, Staphylococcus aureus, and Enterococcus faecalis. Based on the siderophore production, screening conducted Pseudomonas aeruginosa, Staphylococcus aereus, Escherichia coli, Enterococcus faecalis, Rhizobium and Bacillus posses production potential as a result of shiny colonies formation while azotobacter was recorded negative due to non-formation of shiny colonies. Formation of shiny colonies may be due to siderophore production or other chealating compound. Expanding research to isolate and characterize more strains from diverse rhizosphere environments could uncover more efficient and adaptable bacteria, which would broaden their practical applications in agriculture. Field trials should be conducted to evaluate the effectiveness of these isolates under real-world agricultural conditions, particularly to assess their ability to enhance iron uptake and promote plant growth. Keywords: Siderophore, Bacillus subtilis, Pseudomonas aeruginosa, Escherichia coli. Staphylococcus aereus, Rhizobium, Azotobacter, botanical garden, Chrome Azurol Sulfonate assay, chelating compound.
