IN-VITRO ANTIOXIDANT ACTIVITY OF RIPE AND UNRIPE PEEL OF MUSA ACUMINATA(MUSACEAE)ETHANOL EXTRACTS USING 2,2-DIPHENYL-1-PICRYLHYDRAZYL(DPPH)
Abstract
Oxidative stress is one of the triggers of various degenerative diseases and metabolic syndrome.
Antioxidants are compounds that exhibit the activities of neutralizing and scavenging radical
molecules, which induce the process of oxidative reactions in the body. Musa acuminata has
overtime been proven to have antifungal, antibiotic, antimicrobial, anti-diabetic, antiinflammatory and most importantly antioxidant properties amongst its various ethnomedicinal
uses. The present study was aimed at investigating the antioxidant activity of crude ethanol extracts
of ripe and unripe peel of Musa acuminata. The powdered samples of ripe and unripe fruit peel
were extracted with absolute ethanol by cold maceration. Qualitative phytochemical analysis was
done on the extracts with standard procedures. In-vitro antioxidant assay was assessed with 2,2-
diphenyl-1-picrylhydrazyl (DPPH) method. Two-fold serial dilutions were made with ethanol to
obtain concentration of 1000, 500, 250, 125, 62.5, 31.25, 15.63, 7.81 µg/ml of both extracts and
was done in triplicate. The assay mixture contain 1000 µl each of the extract or standard, 1000 µl
of ethanol and 1000 µl of freshly prepared DPPH solution (0.004 mg/ml i.e. 2 mg in 50 ml of
ethanol). The mixture /stock dilutions was incubated at room temperature in the dark for 30min.
The absorbance was determined spectrophotometrically at 517 nm using UV spectrophotometer
(spectrum lab - spm – 752pro). Preliminary phytochemical analysis of the Musa acuminata peel
ethanol extract revealed the presence of glycosides, saponins and flavonoids. The unripe peel (IC

10.92) showed better antioxidant activity than the ripe one ((IC

16.09) and even the Ascorbic
acid (IC

11.79). The present work indicates that the ethanol extract of Musa acuminata exhibits
significant antioxidant activities possibly due to the presence of important phytochemical
constituents and this justify its use in ethnomedicine.
